The xGen® Human mtDNA Research Panel provides complete coverage of the human mitochondrial genome. The panel consists of 138 individually synthesized xGen Lockdown® Probes and delivers reliable coverage of the entire 16 kb mitochondrial genome.

The flexible, modular panel can be used as either a standalone* or spike-in panel for cost-effective and accurate detection of sequence variants in mitochondrial DNA. When added to other xGen Lockdown Panels, ranging in size from <50 kb up to the xGen Exome Research Panel, the mtDNA Research Panel does not affect the high target specificity and coverage uniformity of the original panel.

* The mitochondrial genome comprises a small target space. Therefore, the xGen Human mtDNA Research Panel contains relatively few probes compared to most target capture panels. When used as a standalone panel, expect lower on-target results than when used as a spike in to a larger panel. This reduced performance is typical of small panels in hybridization capture enrichment.

Efficient capture performance of combined panels

The xGen Human mtDNA Research Panel is designed to be used as a spike-in panel to larger capture panels. When used as recommended, the mtDNA Research Panel does not affect target specificity and coverage uniformity of the original panel.

Figure 1. On-target performance of large and small recipient panels is unchanged by addition of xGen® Human mtDNA Research Panel. DNA libraries were prepared from NA12878 genomic DNA (Coriell) using a KAPA Hyper Prep Kit (Kapa Biosystems). Libraries were enriched with the xGen Exome Research Panel (>400,000 probes) or a custom panel comprised of 500 xGen Lockdown® Probes, with and without spike in of the xGen Human ID Panel, using the recommended protocol. The enriched library was sequenced on a MiSeq® System (Illumina). The averages of on-target values (with 150 bp flank) across experiments are shown.
