How do Na+ and Mg2+ concentrations affect the annealing of oligos?
Monovalent and divalent salts like Na+ and Mg2+ are used to stabilize duplex formation by shielding the negatively charged DNA sequence backbone, allowing the two strands to come into closer proximity.
Higher concentrations of these cations will lead to a more stable duplex formation.
